The SASA Calculator measures how much of an uploaded protein structure is accessible to a solvent probe. Surface exposure helps interpret protein interfaces, residue accessibility, and hydrophobic patches that may affect solubility or developability.

The service returns total SASA, per-chain SASA in the assembled structure, isolated-chain SASA, per-residue exposure, and the fraction of exposed residues that are hydrophobic. Multichain inputs also receive a buried-surface estimate.

How SASA Calculator Works

Neurosnap estimates solvent-accessible area with a Shrake–Rupley-style calculation: atom radii are expanded by a solvent probe, points are sampled over each expanded sphere, and points blocked by neighboring atoms do not contribute area. The total is reported in square angstroms.

Per-residue and assembled-chain values are measured with all submitted protein chains present. Isolated-chain values are calculated after selecting each chain alone. Their difference estimates the area buried by assembly; the overall buried SASA is the sum of isolated-chain SASAs minus the assembled SASA. These are geometric exposure measures, not binding free energies. The surface-hydrophobicity fraction counts hydrophobic amino acids among residues with nonzero accessible area.
