Use ESMC Mutation Scoring

Official Neurosnap webserver for accessing ESMC Mutation Scoring online.

Overview

ESMC Mutation Scoring performs zero-shot mutation analysis on a protein sequence using Biohub's ESMC protein language model. The workflow masks each residue one at a time, predicts amino-acid probabilities at every position, and reports per-position entropy, deleterious-fraction summaries, and log-likelihood ratios (LLRs) for all canonical substitutions. Optional submitted variants can be scored as the sum of constituent single-mutant LLRs.

Neurosnap Overview

The ESMC Mutation Scoring online webserver allows anybody with a Neurosnap account to run and access ESMC Mutation Scoring, no downloads required. Information submitted through this webserver is kept confidential and never sold to third parties as detailed by our strong Terms of Use and Privacy Policy.

Features

  • Leave-one-out masked inference across every sequence position with locally hosted ESMC checkpoints.
  • Per-position entropy and deleterious-fraction summaries to identify constrained versus mutation-tolerant sites.
  • Full single-substitution LLR matrix plus ranked top mutations for focused library design.
  • Optional multi-mutant variant scoring from submitted WTposMutant notation.
  • Supports ESMC 300M, 600M, and 6B checkpoints prefetched for offline inference.

Statistics

Neurosnap periodically calculates runtime statistics based on job execution data. These estimates provide a general guideline for how long your job may take, but actual runtimes can vary significantly depending on factors like input size or settings used.
