Overview
CAR-Toner is an AI-driven tool for predicting Chimeric Antigen Receptor (CAR) tonic signaling. It addresses the critical role of tonic signaling in CAR-T cell fitness, where imbalanced signals can lead to poor persistence or exhaustion. By calculating a 'Positively Charged Patch' (PCP) score for the CAR's antigen-binding domain, CAR-Toner quantifies the propensity for CAR clustering and tonic signal induction. Built upon a fine-tuned ESM2 protein language model, it overcomes the slow, manual, and server-dependent limitations of previous methods. The tool provides rapid PCP score calculation for single sequences or large batches.
