How Reverse Complement Works
A reverse complement first pairs each base with its complement, then reverses the sequence to report the opposite strand in the 5′ to 3′ direction. DNA pairs A with T; RNA pairs A with U; C pairs with G in both. Ambiguity codes are complemented to their corresponding IUPAC codes. With automatic sequence typing, each record containing U but no T is treated as RNA, while a record with T or neither base is treated as DNA. Records containing both are invalid.
FASTA records are processed independently, so invalid records are skipped and listed in the warnings while valid records remain available. FASTQ input is validated as a complete read set. Reverse and reverse-complement operations reverse quality strings alongside the sequence, preserving base-to-quality alignment. Select Same as input to retain FASTQ quality scores; FASTA and plain-text output omit them with a warning. The results include a downloadable sequence file and GC percentage calculated from canonical bases in the valid input records.
